how can i improve my threshold in order to calculate the length and width of this THIN LINE using pixel

i'm still new with matlab. I tried threshold, segmentation, but i can,t seem to get a conectivity between my pixel THIN line, so it's hard for me to measure the length and width, please kindly help me plese, this maybe easy for you dears, but really hard for me to do it.

this is what i get when i zoom my threshold, they is no connectivity between the pixel, therefore it.s hard for me to measure the width and length.

Please annotate and upload a picture where you point, in red color, exactly to "my pixel THIN line" because I have no idea what you're talking about. Is there something there in that image that is expected to be just one pixel wide? I'm not seeing it, so please tell me where it is.

Try a bottom hat filter, imbothat().

Sorry for the confiusion guys, the THIN LINE is the line inside the red box, i try measure the width at point A, B , C and D and the exact length of the line inside the red box. So far, i try, enhance contrast, multiple threshold and use pixel to calculate it manually, but the pixel connectivity is not good.
