Compare change of location of items in images

Hi, I am new to matlab and looking for some help on where to begin with this project.

Background: I have a material I am stretching, I am taking images of the material with clear markers on the material, using these as signs of changes between each stretch(image).

I am looking for some type of code in which I can identify the distance between points on the material and then work out the change in the location and distance between each point. Here is an image to demonstrate what I mean by this.

Thank you for any help, even pointers on where to begin.
