HELP WITH Calculatuing area frcation from image contrast

Hi,
I am trying to find area fraction from image contrast. I have no clue how to do it but I know using MATLAB program this can be done. Can you please help me. I am sending one example image. http://postimg.org/image/7nb5h3xqb/

what do you mean from area fraction? Which part of the image would you like to extract?
