improve sharpness of the image

I wanted to know how to improve sharpness of the image? In this way it detects grain boundaries and the size of the nucleus under the microscope. Thank assistants Meir fadida materials engineering student Ben Gurion University

Size of nucleus is not straight forward. U should make a calibration that is, form a ratio between figure and prototype and determine the size of pixel in real units. Then u can isolate one nucleus form figure and u can determine the size by no of pixels forming the nucleus.
