Histogram analysis for images with changing objects

Hi all!! I have done histogram analysis of my images that have mitochondria which change from one area of their image region to another I.e. their shape may change from point a to point b etc. my qualm is I’m not sure what a histogram can give me that would be relevant to what’s happening in the image. If I’m correct in my understanding histogram analysis is the signal of the image so if I want to know more about the mitochondria in the image what can its signal tell me? I’m measuring things like skewness, intensity and kurtosis then I’m putting that into a PCA. Any thoughts about the useful and validity of this? Thanks!

I doubt those histogram stats will help you, but who knows? What is happening in the image? Is the shape something you think is relevant to what is happening, or does it not really matter? Do you have something else that you know, for example temperature, pH, or chemical/drug concentration that is your independent variable and you want to make a model to predict that from various things you can measure in the image such as shape, brightness, area fraction, etc.? If so, you can just measure everything you can think of and use the Regression Learner app on the Apps tab of the tool ribbon to create a model.
